[An improved extraction and nonradioactive thin-layer chromatography detection method of mycolic acid].

Mycolic acids (MAs), i.e. 2-alkyl, 3-hydroxy long-chain fatty acids, are the hallmark of the cell envelope of Mycobacterium tuberculosis and are related with antibiotic resistance and host immune escape. Nowadays, they've become hot target of new anti-tuberculosis drugs. There are two main methods to detect MAs, 14C metabolic labeling thin-layer chromatography (TLC) and liquid chromatograph mass spectrometer (LC-MS). However, the user qualification of 14C or the lack of standards for LC-MS hampered the easy use of this method. TLC is a common way to analyze chemical substance and can be used to analyze MAs. In this study, we used tetrabutylammonium hydroxide and methyl iodide to hydrolyze and formylate MAs from mycobacterium cell wall. Subsequently, we used diethyl ether to extract methyl mycolate. By this method, we can easily extract and analyze MA in regular biological labs. The results demonstrated that this method could be used to compare MAs of different mycobacterium in different growth phases, MAs of mycobacteria treated by anti-tuberculosis drugs or MAs of mycobacterium mutants. Therefore, we can use this method as an initial validation for the changes of MAs in researches such as new drug screening without using radioisotope or when the standards are not available.

The Surface-to-Atmosphere GHG Fluxes in Rewetted and Permanently Flooded Former Peat Extraction Areas Compared to Pristine Peatland in Hemiboreal Latvia

When it comes to greenhouse gas (GHG) reduction, the role of water tables in former peat extraction areas has received considerable interest in recent decades. This study analysed the carbon dioxide (CO2), methane (CH4) and nitrous oxide (N2O) surface-to-atmosphere fluxes from a rewetted and permanently flooded former peat extraction areas in comparison to pristine peatland in hemiboreal Latvia. Measurements of GHG fluxes combined gas sampling using a closed-chamber (opaque) method with the gas chromatography detection method. Among the studied land-use types, the highest annualised CO2 fluxes (soil heterotrophic and autotrophic respiration) were recorded in rewetted former peat extraction areas with restored vegetation and in undisturbed peatland (4.10 ± 0.21 and 3.45 ± 0.21 t CO2-C ha−1 yr−1, respectively), with the lowest in flooded former peat extraction areas (0.55 ± 0.05 t CO2-C ha−1 yr−1); temperature and groundwater level were found to be significant influencing factors. The highest annualised CH4 fluxes were recorded in undisturbed peatland (562.4 ± 155.8 kg CH4-C ha−1 yr−1), followed by about two-fold and ~20-fold smaller CH4 fluxes in flooded and rewetted areas, respectively. N2O fluxes were negligible in all the studied land-use types, with the highest N2O fluxes in undisturbed peatland (0.66 ± 0.41 kg N2O-N ha−1 yr−1).

Green construction of hydroxyl-functionalized magnetic porous organic framework for effective extraction of triazine herbicides from environmental water and watermelon juice samples

Triazine herbicides have been widely detected in water resources and food, which poses a potential hazard to both ecosystem and human health. Due to their high polarity, conventional adsorbents have limitations for their extractions. Herein, for the effective magnetic extraction of triazine herbicides, a novel and effective magnetic adsorbent was prepared with a satisfactory extraction performance. In the experiments, five porous organic frameworks (POFs) with hydroxyl functional groups were synthesized by diazo-coupling reactions in aqueous solution with β-cyclodextrin (β-CD) as a green monomer. After evaluation of the five POFs, the DDM-CD-POF, which was synthesized with 4′4-diaminodiphenylmethane (DDM) and β-CD, showed the largest specific surface area and the best adsorption capacity for the five triazine herbicides. Then, it was magnetized by introducing Fe3O4@SiO2 into it to prepare a magnetic adsorbent (M-DDM-CD-POF) to facilitate separation and recycling. Finally, the M-DDM-CD-POF-based magnetic solid-phase extraction in combination with high performance liquid chromatographic detection method was established for the quantitative determination of the triazine herbicides in environmental water and watermelon juice samples. The current strategy showed low limits of detection of 0.03–0.11 ng mL−1 for environmental water and 0.07–0.22 ng mL−1 for watermelon juice sample. The method recoveries for spiked samples ranged from 84.0% to 113.0% with the relative standard deviations ≤8.8%. This work provides a new approach for the detection of the triazine herbicides with good application prospect.

Recent Advances in the Development and Characterization of Electrochemical and Electrical Biosensors for Small Molecule Neurotransmitters.

Neurotransmitters act as chemical messengers, determining human physiological and psychological function, and abnormal levels of neurotransmitters are related to conditions such as Parkinson's and Alzheimer's disease. Biologically and clinically relevant concentrations of neurotransmitters are usually very low (nM), so electrochemical and electronic sensors for neurotransmitter detection play an important role in achieving sensitive and selective detection. Additionally, these sensors have the distinct advantage to potentially be wireless, miniaturized, and multichannel, providing remarkable opportunities for implantable, long-term sensing capabilities unachievable by spectroscopic or chromatographic detection methods. In this article, we will focus on advances in the development and characterization of electrochemical and electronic sensors for neurotransmitters during the last five years, identifying how the field is progressing as well as critical knowledge gaps for sensor researchers.

Synthesis of spherical amine-functionalized silica molecular sieve and application as selective adsorbents for aromatic hydrocarbons analysis

In this work, spherical amine-functionalized mesoporous silica (Sp-Amino-Si) molecular sieve was fabricated by an acid hydrothermal method using 3-aminopropyltriethoxysilane as a modifier and toluene as the solvent. The results of scanning electron microscopy, Barrett-Joyner-Halenda pore size distribution, Fourier transform-infrared spectroscopy, and low-angle X-ray diffraction patterns revealed that the Sp-Amino-Si was monodispersed microspheres with uniform and controllable pore sizes sharing two-dimensional hexagonal P6mm symmetry and a large number of amino groups. The Sp-Amino-Si molecular sieve was verified to be suitable for the selective separation of aromatic hydrocarbons (AMHs) containing two fused benzene rings among other compounds from environmental water matrix. Under the optimized conditions, the Sp-Amino-Si exhibited much higher adsorption efficiency for AMHs (above 88.19%), comparing to the original SBA-15 (below 44.52%). The maximum adsorption capacities for naphthalene, methyl naphthalene, 2-methyl naphthalene, and acenaphthene were 477.1, 1006.5, 893.3, and 1253.2 µg·g−1, respectively. The high selectivity of Sp-Amino-Si for the four AMHs attributed to the size sieving and p-π interactions between the lone pair electrons of amine groups and the π-electrons in AMHs molecules. The Sp-Amino-Si based solid-phase extraction coupled with high performance liquid chromatography detection method was constructed for the analysis of the four AMHs, and the recoveries of spiked AMHs ranged from 89.67% to 112.74% with a relative standard deviation lower than 5.75%. This study provides some inspiration and a reliable method for AMHs separation, removal, and analysis.

Development of a thin‐layer chromatographic method for detection of lipstatin produced by Streptomyces toxytricini

AbstractLipstatin or its saturated analog (orlistat), being the irreversible inhibitor of intestinal lipase, is widely used for the treatment of obesity. The bacterium Streptomyces toxytricini is the main source for the production of lipstatin/orlistat. There are continuous attempts to increase the production of lipstatin in S. toxytricini. For optimization of best conditions/strains for lipstatin production, there are requirements for a fast, simple, and reliable method to detect/estimate lipstatin. At present, highly sophisticated methods such as high‐performance liquid chromatography, mass spectrometry, and so forth are available for the detection and estimation of lipstatin/orlistat. These methods are very costly, time‐consuming, and require state of the art facility. Here we report a simple, fast, cost‐effective method based on thin‐layer chromatography for the detection of lipstatin in S. toxytricini. The optimized mobile phase was acetone: ethanol in a ratio of 3:7. Development of thin‐layer chromatography was done by ρ‐anisaldehyde staining that showed the pink color to the silica plate and greenish‐blue color to the lipstatin/orlistat drug. Conclusively, here we mention a simple and reproducible method of detection of orlistat and lipstatin samples extracted from S. toxytricini.

Rapid detection of SARS-CoV-2: The gradual boom of lateral flow immunoassay.

Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) is still in an epidemic situation, which poses a serious threat to the safety of people and property. Rapid diagnosis and isolation of infected individuals are one of the important methods to control virus transmission. Existing lateral flow immunoassay techniques have the advantages of rapid, sensitive, and easy operation, and some new options have emerged with the continuous development of nanotechnology. Such as lateral flow immunoassay test strips based on colorimetric-fluorescent dual-mode and gold nanoparticles, Surface Enhanced Raman Scattering, etc., these technologies have played an important role in the rapid diagnosis of COVID-19. In this paper, we summarize the current research progress of lateral flow immunoassay in the field of Severe Acute Respiratory Syndrome Coronavirus 2 infection diagnosis, analyze the performance of Severe Acute Respiratory Syndrome Coronavirus 2 lateral flow immunoassay products, review the advantages and limitations of different detection methods and markers, and then explore the competitive CRISPR-based nucleic acid chromatography detection method. This method combines the advantages of gene editing and lateral flow immunoassay and can achieve rapid and highly sensitive lateral flow immunoassay detection of target nucleic acids, which is expected to be the most representative method for community and clinical point-of-care testing. We hope that researchers will be inspired by this review and strive to solve the problems in the design of highly sensitive targets, the selection of detection methods, and the enhancement of CRISPR technology, to truly achieve rapid, sensitive, convenient, and specific detection of novel coronaviruses, thus promoting the development of novel coronavirus diagnosis and contributing our modest contribution to the world's fight against epidemics.

Thin Layer Chromatographic Method for Detection of Conventional Drug Adulterants in Herbal Products

Commercially available conventional drugs have been used to adulterate herbal products. Considering the rapid growth of herbal products’ market, it is essential to screen herbal products for the presence of conventional drugs. Simple analytical methods are needed for the rapid screening of conventional drugs that are likely to be adulterated in herbal products. Thin layer chromatography (TLC) methods for screening twelve conventional drugs in herbal products have been developed and applied. The analytes were extracted from herbal products using acetonitrile:methanol:acetic acid:water (4:4:1:1, v/v). Solvent mixture of dichloromethane:ethyl acetate:methanol (75:15:10, v/v) separated well trimethoprim, sildenafil, paracetamol, and sulfamethoxazole while pyrimethamine, metronidazole, and sulfadoxine were well separated by dichloromethane:ethyl acetate:methanol (77.5:12.5:10, v/v). In addition, acetyl salicylic acid, ibuprofen, diclofenac, quinine, and lumefantrine were well separated by ethyl acetate:methanol:30% ammonia (75:22.5:2.5, v/v). Chromatographic separations were found to be highly reproducible, and more than 10 samples can be analysed in one run. The method was applied in the screening of 229 herbal products. Consequently, 24.0% of the samples contained one adulterant, while 21.4% contained at least two adulterants. All conventional drugs detected in herbal products were not mentioned on the labels and therefore the consumers are kept unaware of their side effects and health problems. Further studies for confirming and quantitatively determining the adulterants in a wide range of products as well as a systematic toxicological analysis of the adulterants in herbal products are recommended.

Study on Integrated Pharmacokinetics of the Component-Based Chinese Medicine of Ginkgo biloba Leaves Based on Nanocrystalline Solid Dispersion Technology

BackgroundTo improve the dissolution and bioavailability of the component-based Chinese medicine of Ginkgo biloba leaves (GBCCM), a novel nanocrystalline solid dispersion of GBCCM (GBCCM NC-SD) was first prepared.MethodsGBCCM mainly containing high pure flavonoid aglycones (FAs) and terpenoid lactones (TLs) was used as the model drug. PVP K30 and SDS were used as solubilizers, combined stabilizers and carriers, and GBCCM NC-SD was prepared by high-pressure homogenization combined with freeze-dryer. Morphology and crystal characteristic of GBCCM NC-SD were analyzed. The dissolution and bioavailability evaluation were performed to investigate the feasibility of GBCCM NC-SD by in vitro dissolution and in vivo integrated pharmacokinetic models.ResultsAfter homogenizing for 30 cycles under the pressure of 650 bar and freeze-drying, GBCCM NC-SD with uniform quality would be obtained. The particle size, PDI and zeta potential were found to be 335.9 ± 32.8 nm, 0.29 ± 0.02 and −28.4 ± 0.7 mV respectively. Based on charged aerosol detector (CAD) technology, a new chromatographic method for simultaneous detection of eight components in GBCCM was developed. In vitro drug release study showed that the cumulative dissolution of FAs and TLs in GBCCM NC-SD increased from 12.77% to 52.92% (P < 0.01) and 90.91% to 99.21% (P < 0.05) respectively. In comparison with physical mixture of GBCCM and stabilizer (PM), the integrated pharmacokinetics AUC0-t of FAs and TLs in GBCCM NC-SD were significantly increased (P < 0.05), and the T1/2 of TLs was also significantly prolonged (P < 0.05).ConclusionThis study demonstrated that novel GBCCM NC-SD was prepared using Polyvinylpyrrolidone K30 (PVP K30) and Sodium dodecyl sulfate (SDS) as a synergetic stabilizer and also provided a feasible way to improve the dissolution and oral bioavailability of poorly soluble candidate antihypertensive drugs.

HPLC-DAD assay of phenols profile in Antennaria dioica (L.) Gaertn

Antennaria dioica (Asteraceae family) – is a perennial herb, commonly found in dry grasslands and sandy or stony places from Eurasian areas. It is known in traditional medicine as antioxidant, diuretic, choleretic and anti-inflammatory remedy. This species should be reconsidered as possible sources of phenols, mainly flavonoids and hydroxycinnamic acids. Thus, the aim of this study was to validate a chromatographic method for detection of phenols and their identification in A. dioica herb. HPLC-DAD method was evaluated in terms of linearity, precision, repeatability, accuracy, LOD and LOQ. The calibration curves of thirteen reference substances were linear (R2 &amp;gt; 0.99) over the range of 5–400 µg/mL, the LODs and the LOQs were in the range of 0.1–0.3 µg/mL and 0.2–1.0 µg/mL, respectively. During HPLC-DAD assay two flavones – luteolin, apigenin; flavonol – quercetin and three its glycosides – rutin, hyperoside and isoquercitrin; coumarins: coumarin and umbelliferone; five hydroxycinnamic acids – chlorogenic, caffeic, p-coumaric, trans-ferulic and rosmarinic were identified in A. dioica herb. This phytochemical study of A. dioica confirms that this plant material is a rich source of phenolic compounds.

The optimal combination of Nile red identification, colony polymerase chain reaction, and gas chromatography detection methods in screening for polyhydroxyalkanoicate-producing bacteria.

The study devised a detection process combining Nile red-containing media, polymerase chain reaction (PCR), and gas chromatography (GC) to evaluate the possibility of microbes becoming polyhydroxyalkanoate (PHA) producers. The Nile red and PCR detection steps of designating PHA producers had true positive rates of 39.4% and 40%, respectively, and the use of GC analysis as the final step yielded accurate results for the production types and yields of PHAs. When the number of screening samples was up to 102, connecting all three inspection methods in tandem generated economic benefits. When up to 105 samples were screened, the use of all three detection methods reduced the cost to 3% of the cost and the time consumed 6% of using just Nile red plus GC or PCR plus GC. However, when the sum of samples exceeded 108, the cost of combining the three methods exceeds 1 million US dollars and was excessive; here, the combination of Nile red plus PCR could be considered, even though the true positive rate was only 30.7%.

HPLC-FLD analysis of amino acids content in Chrysanthemum morifolium

Chrysanthemum morifolium (Asteraceae family) have long been used as a tonic, antioxidant, antipyretic, analgesic, sedative, antitumor, neuroprotector, hepatoprotector and cardioprotector agent. This species should be reconsidered as possible sources of many biocompounds, especially amino acids. Thus, the aim of this study was to validate the chromatographic method for detection of amino acids and their identification in flowers and leaves of Ch. morifolium of variant Pectoral. HPLC-FLD method was evaluated in terms of linearity, precision, repeatability, accuracy, LOD and LOQ. The calibration curves of all analytical standards of amino acids were linear (R2 &amp;gt; 0.99) over the range of 0.015–0.625 μmol/mL, the LODs and the LOQs were in the range of 0.001–0.096 µg/mL and 0.004–0.321 µg/mL, respectively. During the HPLC-FLD assay ten amino acids in free form and fifteen amino acids after hydrolysis in Ch. morifolium flowers were identified. Besides, twelve amino acids were detected in free form and fourteen amino acids after hydrolysis in Ch. morifolium leaves. The results of HPLC-FLD analysis showed that the predominant amino acid was L-proline in both types of herbal raw materials. Its total content was 31.67±0.02 μg/mg in Ch. morifolium flowers and 18.56±0.02 μg/mg in Ch. morifolium leaves. This phytochemical study confirms that flowers and leaves of Ch. morifolium (Pectoral) are rich sources of amino acids and can exhibit a wide range of pharmacological activities.

Method Development and Validation of Gas Chromatography Methods using Nitrogen Phosphorus Detector (GC-NPD) and Isotope Dilution Triple Quadrupole Mass Spectrometry (GC-IDMS/MS) for the Determination of Chlorpyrifos in Mango

Pesticides residue determination in fruits like chlorpyrifos is important due to its health effects on humans. Thus, there is a need for sensitive and accurate methods for better quantification. In this study, the performance of two gas chromatographic detection methods, nitrogen phosphorus detector (GC-NPD) and isotope dilution triple quadrupole mass spectrometer (GC-IDMS/MS), for the determination of chlorpyrifos in mango was evaluated to be used for the development of candidate reference material. Other than liquid-liquid extraction (LLE), Quick, Easy, Cheap, Efficient, Rugged and Safe (QuEChERS) sample extraction and dispersive solid phase extraction (dSPE) clean-up procedures were optimized to extract chlorpyrifos from the sample matrix. Comprehensive method validations were performed in GC-NPD and GC-IDMS/MS with HP-5 capillary column to establish linearity, the limit of detection (LOD), the limit of quantification (LOQ), accuracy, repeatability, and intermediate precision using freeze-dried mango samples. The LOD and LOQ were 2.35 and 3.47 µg kg-1  for GC-NPD, and 1.23 and 1.85 µg kg-1 for GC-IDMS/MS, respectively. Matrix-matched calibration curves showed excellent linearity of r2=0.999 for both GC-NPD and GC-IDMS/MS protocols. Acceptable repeatability was obtained for three spike concentrations of 10, 30, and 60 µg kg-1 (n=10) and intermediate precision for 0.5-, 1- and 3-months in the same three-spike concentrations (n=3) expressed as relative standard deviation (RSD) ranging from 0.34-5.61 % for GC-NPD and 0.46-4.22 % for GC-IDMS/MS. Satisfactory mean recoveries (n=10) were achieved: 88.48-109.81% in GC-NPD and 97.61-108.40% in GC-IDMS/MS. GC-IDMS/MS and GC-NPD methods were fit for the purpose of quantifying chlorpyrifos in Philippine mango accurately with the use of isotope dilution technique and traceability to international standards by the application of gravimetric sample preparation.

Determination of streptomycin and doxycycline using LC/MS towards an effective treatment against an experimental Brucella abortus infection in mice

Herein we described a versatile liquid chromatographic method for detection and quantification of the total levels of two antimicrobials [i.e., streptomycin (STM) and doxycycline (DOX)], in mice plasma and selected tissues, with the aid of a single quadrupole as a detection method. The method included a few sample preparation steps, including freeze-drying and in situ triphasic solvent-assisted defatting, precipitation, and extraction, allowing easy and fast tissue sample processing and avoiding analyte loss. Using a murine model, we demonstrated that mass spectrometry detects simultaneously and with high specificity two of the most widespread antimicrobials used against Brucellosis. An accurate [recoveries varied from 75.23 (bone marrow) to 101.33% (liver)] and sensitive (LoD in the ng g−1 range) method to assess STM and DOX in murine tissue, including subtherapeutic and therapeutic doses of the antimicrobials, was achieved. This validated method can be successfully used to monitor the depletion of STM and DOX in several mice tissues and plasma during metabolism after administration.

Identification of a New Badnavirus in the Chinaberry (Melia azedarach) Tree and Establishment of a LAMP-LFD Assay for Its Rapid and Visual Detection.

The Chinaberry tree, a member of the Meliaceae family, is cultivated in China for use in traditional medicines. In 2020, Chinaberry trees with leaf deformation symptoms were found in Hangzhou, Zhejiang province, China. In order to identify possible pathogenic viruses, a symptomatic sample was subjected to deep sequencing of small interfering RNAs. Assembly of the resulting sequences led to the identification of a novel badnavirus, provisionally designated Chinaberry tree badnavirus 1 (ChTBV1). With the recent development of China’s seedling industry and increasing online shopping platforms, the risk of tree virus transmission has increased substantially. Therefore, it is important to detect the occurrence of ChTBV1 to ensure the safety of the Chinaberry tree seedling industry. Here, we describe the development and validation of a sensitive and robust method relying on a loop-mediated isothermal amplification (LAMP) assay, targeting a 197 nt region, to detect ChTBV1 from Chinaberry tree leaves. The LAMP assay was also adapted for rapid visualization of results by a lateral flow dipstick chromatographic detection method.

A systematic review on sensing techniques for drug- facilitated sexual assaults (DFSA) monitoring

The use of recreational substances in drug-facilitated sexual assaults (DFSA) has drawn attention during the last few decades and has become a significant problem worldwide. Public awareness in context to DFSA is restricted, therefore early recognition and management of such crucial cases became problems for non-forensic professionals and on-spot investigators. Although forensic laboratories are using conventional chromatographic and spectroscopic methods for DFSA drug detection, expertise requirement in result analysis and time consuming detection method always calls for rapid and on-spot sensing platform. Therefore, numerous researches have been focused on developing sensors for sensitive DFSA associated drug detection. In this review, we summarize previous technological advancements in sensor development for on-spot DFSA drug detection and futuristic potential approaches for rapid analytical methods. Our study focuses on sensing of DFSA drugs, namely Rohypnol® (flunitrazepam), Gamma Hydroxy Butyrate (GHB) and ketamine followed by potential limitations for real life implementations of the sensors. In conclusion, we summarizeare summarizing attempts made in sensing technonolgy development for on-spot and rapid DFSA drugs detection. To the best of our knowledge,to date no effort has been made in this direction, hence our study may provide detailed insight of forensic science and analytical chemistry. We believe it will motivate the scientific community to make additional effort in this emerging interdisciplinary field for DFSA associated drugs detection.

DETECTION OF FUNGICIDE RESIDUES IN SOME FRUITS AND VEGETABLES BY CHROMATOGRAPHIC METHODS

The use of pesticides is a common practice in modern agriculture. To increase and to ensure the security in people nutrition, two comparative chromatographic methods (GC-MS and HPLC-DAD) for fungicide residues detection and monitoring in some fruits and vegetables were tested. Some vegetables, eggplants, cucumbers, red potatoes, white potatoes, red peppers and fruits, plums, apples, lemons, grapes, clementines, were analyzed for fungicide residues quantification. The presence of Tebuconazole, Boscalid, Iprodione and Imazalil was followed and quantified in the tested samples. These analyses are important for environment and end-consumers protection. Generaly, both techniques are very sensitive and selective for the analysis of pesticides at low concentrations. GC-MS presents better linearity characteristics for Tebuconazole and Imazalil, but HPLC-DAD was the most rapid method. The tested products do not contain large fungicides quantities.

The Implications of Chromatographically Screening Medical Products for Organic Leachables Down to the Analytical Evaluation Threshold Adjusted for Response Factor Variation.

A drug product is chromatographically screened for organic leachables, derived from the product's packaging system, as leachables might adversely impact the health of a patient to whom the drug product is administered. Similarly, medical device and packaging system extracts are chromatographically screened for organic extractables as probable leachables. To be protective of patient health, the screening methods must produce recognizable responses for all potentially unsafe substances. To be efficient, the screening methods should provide a means of differentiating between the responses linked to likely to be safe substances and to potentially unsafe substances. The analytical evaluation threshold (AET) was established as a means of differentiating chromatographic peaks, based on concentration, that are unlikely to be unsafe (and thus do not need safety assessment) and that are possibly unsafe (and thus require safety assessment). Thus, the AET manages the competing objectives of protection and efficiency. Although the AET is based on concentration, it is applied based on response. As no chromatographic detection method applied to extractables and leachables screening produces a uniform response to all potential analytes (thus, the magnitude of the response differs across analytes), the objectives of protection or efficiency can be compromised by false negatives and positives. To ensure protection at the expense of efficiency, the AET can be adjusted to address response variation. This article addresses the practical issue that the protectiveness of the AET is affected both by response factor bias and variation and thus correction for only variation is incomplete and ineffective. Chromatographic Methods for Detection and Quantification of Carbendazim in Food.

Carbendazim (CBZ), which is a fungicide widely used for the management of plant diseases, has been detected in a number of food products. The negative effects of CBZ to human health have stimulated the reduction of the maximum residue limits (MRLs), and subsequently the development of reliable and sensitive detection methods. Here, we are reviewing for the first time all reported chromatographic methods for the detection and quantification of CBZ in food. Several techniques, including liquid chromatography (LC), thin layer chromatography (TLC), micellar electrokinetic chromatography (MEKC), and supercritical fluid chromatography (SFC), were used for the separation and detection of CBZ, showing diverse characteristics and sensitivity. Some methods allowed the specific determination of CBZ, whereas other methods were successfully applied for the simultaneous quantification of a huge number of pesticides. Most reported methods showed limits of detection (LOD) and quantification (LOQ) lower than the MRLs. Relevant efforts in the field have been directed toward the simplification and optimization of the extraction steps prior to the chromatographic separation to increase the recovery and reduce the matrix effects. In this Review, the matrices, extraction procedures, and separation and detection parameters are detailed and compared in order to provide new insights on the development of new reliable methods for the detection of CBZ in food.
